Probably the most challenging part is not the culture itself but the selection (i.e. identification and picking; combination of imaging software and robotics) of the best reprogrammed colonies prior to their expansion. There has been some progress (e.g. https://www.nature.com/articles/s41598-017-13680-x) but still lacking a complete system capable of doing just that.
